PAMPA Explorer™ gives you access to pION's leading edge PAMPA technology, including the patented Double-Sink™ technology.  It includes a license for our PAMPA Explorer software and detailed instructions for running the PAMPA assay.

Double-Sink™
Our patented Double-Sink model uses pH gradient and an acceptor sink to mimic in vivo passive transport. This method correlates well with human absorption.

Gut-Box
Mechanical stirring apparatus for use with the PAMPA assay

The Gut-Box™ is an important accessory for pION's PAMPA system. Designed with a built-in stirring mechanism, it is calibrated to specific speeds so that the PAMPA assay mimics the expected thickness of the aqueous boundary layer. The Gut-Box also provides controls for humidity, oxygen, and carbon dioxide.

When the Gut-Box™ is used with the Double-Sink™ PAMPA assay, PAMPA Evolution™ and PAMPA Explorer™, permeability assays can be done in less than 10 minutes.
µSOL Explorer

µDISS ProfilerPLUS™
Small Volume Dissolution and Solubility Apparatus

In response to the increasing interest in characterizing intrinsic dissolution in early development where the active pharmaceutical ingredient is not available in large quantities, the µDISS ProfilerPLUS™ has been developed for small masses and volumes.

The µDISS Command software allows for easy method development. The µDISS ProfilerPLUS™ can be used to:

  • Rank order intrinsic dissolution
  • Detect polymorphic changes
  • Follow stability profiles
  • Determine equilibrium solubility
Gemini Profiler™

The Gemini Profiler™ is a semi-automated, state-of-the-art potentiometric analyzer for measuring pKa.  The instrument uses the potentiometric titration method developed by Alex Avdeef (pION's CEO & CSO).  The Gemini Profiler™ advantages include:

  • Determnes pKa in the presence of some precipitation, since the instrument can correct for intrinsic solubility.
  • The Gemini Profiler™ does not need to perform separate blank titrations.  The "ABC" electrode parameters can be determined in situ, at the same time that pKa values are determined.
  • Any co-solvent can be used, including the water-miscible solvents polyethylene glycol, bile salts, and methylpyrrolidone.  It also can determine co-solvent pKa < 2 and pKa > 12.
  • Determines log P and log D of ionized compounds.
  • Determines intrinsic solubility and solubility as a function of pH (1-13, increments of 0.1)

Drug Discovery

With as little as 100 micrograms of sample, the Gemini Profiler™ delivers a complete solubility-pH profile. This allows you to make solubility an integral part of your optimization program at earlier stages of drug discovery, by allowing you to experimentally "calibrate" your high-throughput solubility prediction calculations. The result is earlier prediction of oral absorption.  Determination of pKa can be done separately or in conjunction with solubility measurement.

Drug Development

For the same effort as a manual single-pH solubility measurement, the Gemini Profiler™ determines the whole pH range, automatically. Not only aqueous solubilities, but also solubilities in DMSO, methanol and acetonitrile. Following guidelines consistent with the BioPharmaceutics Classification System, dose predicting is made easier. Solubilities less than 5 ng/mL to 50 mg/mL are possible.  You will not want to do another shake-flask solubility determination. pKas of sparingly soluble compounds are easily determined using a co-solvent extrapolation method.  In addition, compounds with multiple or overlapping pKas are not a problem for the Gemini Profiler™.
